Asante Kotoko, the last FA Cup holders will kick off the defence of their trophy with a home tie against Danbort FC in the Round of 64.
The Porcupine Warriors, who won the last Cup decade ago, were drawn against the Accra-based Division Two side following Friday's live draw for the Round of 64 of the MTN FA Cup.
Ties for the one-off games will be played on the weekends of 21-22 and 28-29 January.
Other intriguing ties see Hearts of Oak host Pure Joy and the all-premiership clashes - Berekum Chelsea take on AshantiGold, Dwarfs battle Aduana, Medeama play Real Tamale United.
All 16 Premier League sides were entered into the draw which also includes 17 Division Two sides and 31 clubs from the Division One League.
The competition has already recorded casualties but as it reaches the business stage, the possibility of having more upsets looms with some lower tier clubs drawing Premiership sides.
