Defending champions Asante Kotoko will host Asokwa Deportivo at the Len Clay stadium whilst Hearts of Oak will travel to play Young Wise FC in the MTN FA Cup round of 16.
The draw which took place in Accra on Tuesday saw nine Premier League teams pitted against seven lower league teams.
The Disciplinary Committee of the Ghana Football Association is however expected to make a decision on the match between Legon Cities and Phar Rangers, with the latter being investigated by the FA.
The 16 teams were grouped into two regional zones for the draw; Northern and Southern.
There were some mouthwatering ties including an all Premier League affair between Aduana Stars and AshantiGold, and then Medeama who play Great Olympics.
